Autophagy is an intracellular degradation system that delivers cytoplasmic constituents to the lysosome. Despite its simplicity, recent progress has demonstrated that autophagy plays a wide variety of physiological and pathophysiological roles, which are sometimes complex.

2016諾貝爾生理暨醫學獎頒給日本科學家大隅良典!
以表彰他對細胞的自我吞噬(autophagy)的研究。
細胞為什麼要自我吞噬?
因為要移除細胞內的老舊胞器,
或者打擊入侵的微生物,
算是一種新舊汰換與抵禦外侮的機制。
怎麼進行?
當細胞內的胞器過了生命周期,
便在細胞內形成自噬體(autophagosome),
那是宛如一個塑膠袋的東西,
將不要的胞器包裹起來,
再跟細胞內的溶小體(lysosome)融合,
讓老舊的胞器被溶小體內的水解酶分解,
然後大功告成。
自我吞噬的現象在1960年代就已發現,
但直到1993年,
才由當時在東京大學擔任助理教授的大隅良典,
藉由酵母菌的實驗而發現作用機制,
並找出了多達十五個相關基因,
他當時四十八歲。
大隅良典想到,
如果酵母菌存在自我吞噬,
那麼抑制空泡(類似溶小體)的作用,
將會導致被圈住的細胞質堆積,
因為無法被溶解,
於是他培養出欠缺空泡蛋白酶的酵母菌,
證實了這樣的想法。
大隅良典為什麼會想到用酵母菌做實驗?
又為什麼認定酵母菌也有自體吞噬?
是不是諾貝爾等級的科學家,
就決定在這個眉角。
他是第四位獲得諾貝爾生理暨醫學獎的日本人,
這幾年日本科學家接連獲得該獎項,
相當不簡單。
大隅良典是東京大學博士,
1970年代曾到美國做博士後研究幾年,
其他時間都待在日本。
